Pharmacologically the compound activates three receptors: GLP-1, GIP, and glucagon. GLP-1 and GIP signaling contribute to glucose-dependent insulin release, delayed gastric emptying, and reduced appetite, while glucagon receptor activation is associated with increased energy expenditure and hepatic fat oxidation. The single-molecule design is intended to keep these activities in one peptide rather than combining separate agents. Relative activity at each receptor differs, and the balance between them is a central question in interpretation. The glucagon component is partly offset by incretin-mediated insulin secretion, an interaction that remains incompletely characterized.
Development has progressed from single- and multiple-ascending-dose studies in healthy volunteers into larger randomized trials in adults with obesity, type 2 diabetes, and fatty liver disease. Early reports describe dose-dependent reductions in body weight and improvements in glycemic markers over treatment periods of several months. Whether the glucagon arm adds tolerability cost without added benefit is still debated. Long-term cardiovascular outcomes, effects after treatment stops, and performance in older or comorbid populations are open questions rather than settled findings. Approval status may change, so the current investigational label should be confirmed against regulatory sources.
Retatrutide is an investigational synthetic peptide developed under the code LY3437943, with a backbone derived from glucose-dependent insulinotropic polypeptide. Several non-proteinogenic residues, including alpha-aminoisobutyric acid, appear in that backbone, and a fatty diacid side chain attached through a linker extends circulation time. The molecule carries roughly thirty-nine amino acid units and a total mass near 4.7 kilodaltons. Administration is by subcutaneous injection once weekly. Published work uses both the name retatrutide and the code LY3437943.
Characterization panels may add amino acid analysis for compositional confirmation, circular dichroism for secondary structure in solution, and light scattering for aggregation tendency. Aggregation is a central concern for peptides bearing hydrophobic side chains, since it can lower measured potency and complicate accurate dosing. Stability studies examine temperature, humidity, pH, and light exposure over defined intervals, reporting the percentage of intact peptide remaining. Results depend strongly on the assay used, so comparing values across studies requires matching method details.
Peptide content and purity are commonly measured by reversed-phase high-performance liquid chromatography with ultraviolet detection, using gradient elution over a C18 column. Identity is confirmed by mass spectrometry, because the theoretical monoisotopic mass allows unambiguous assignment of the main component. Impurity profiling resolves deletion sequences, oxidized residues, and truncated fragments. Since the molecule carries a lipophilic side chain, mobile phases often include ion-pairing agents and organic modifiers to keep peaks symmetric.
Mechanistic proposals link each receptor to a different physiological role. Activation of the glucose-dependent insulinotropic polypeptide and glucagon-like peptide-1 receptors is associated with reduced appetite, slower gastric emptying, and glucose-dependent insulin release. Glucagon receptor signaling, by contrast, is associated with increased energy expenditure and altered lipid handling, though it can also raise blood glucose. The design intent is to balance these contributions so that weight reduction is enhanced without unacceptable glycemic trade-offs. How well that balance holds across individuals is not fully resolved.
Published information places retatrutide in clinical development rather than on the market as an approved therapy. Early-stage and mid-stage trials have examined tolerability and changes in body weight, and larger studies continue to report results over time. Open questions include the durability of effects after treatment stops, the composition of weight lost, and cardiovascular outcomes over long periods. Statements about definitive benefit should therefore be treated as provisional. Regulatory status varies by jurisdiction and changes as applications are reviewed.
== Treatment == Currently, there is no cure for Urbach–Wiethe disease, although there are some ways to individually treat many of its symptoms. There has been some success with oral dimethyl sulfoxide (DMSO) and intralesional heparin, but this is not true in all cases. D-penicillamine has also shown promise, but has yet to have been used extensively. There are also some reports of patients being treated with etretinate, a drug typically prescribed to treat psoriasis. In some cases, calcifications in the brain can lead to abnormal electrical activity among neurons. Some patients are given anti-seizure medication to help deal with these abnormalities. Tracheostomy is often used to relieve upper respiratory tract infections. Carbon dioxide laser surgery of thickened vocal cords and beaded eyelid papules have improved these symptoms for patients. The discovery of the mutations of the ECM1 gene has opened the possibility of gene therapy or a recombinant EMC1 protein for Urbach–Wiethe disease treatment, but neither of these two options are currently available.
=== 2010s onwards === The closure of the Delta hub resulted in several changes at the Memphis airport. Passenger traffic declined from eleven million in 2007 to four million in 2017. The terminal, with its three concourses and unoccupied gates, was too large. In 2014, the Memphis–Shelby County Airport Authority announced a renovation of the airport that would involve modernizing Concourse B and mothballing the other two concourses. On the other hand, airfares dropped and new low-cost airlines entered the market, such as Southwest Airlines. The airport continued to see a high level of cargo activity due to the FedEx hub. As of 2018, Memphis was the second-busiest cargo airport in the world. The refurbishment of Concourse B was completed in February 2022. The building features wider hallways and higher ceilings. The southwest wing of the concourse was not renovated because of cost and a lack of need for the additional gates. Work commenced on an expansion of the landside portion of the terminal in 2024. The program includes tearing down Concourse A to make room for an administrative and baggage-inspection building. Following the death of Frederick W. Smith, founder of FedEx, in June 2025, the airport authority decided to rename the facility in his honor due to his contributions to the city of Memphis and the airport in particular. The Federal Aviation Administration was working on implementing the change as of December 2025. On August 11, 2026, the airport was officially renamed to Frederick W. Smith International Airport.
In 2000, it acquired Brazilian appliance maker Multibrás, owner of the brands Brastemp and Consul, including its stake in Embraco. In 2001, Inglis Ltd. changed its name to Whirlpool Canada. Whirlpool continues to market Inglis appliances to this day.
=== Complications === GDM poses a risk to the mother and child. This risk is largely related to uncontrolled blood glucose levels and their consequences. The risk increases with higher blood glucose levels. Treatment resulting in better control of these levels can reduce some of the risks of GDM considerably. Having GDM can lead to mental health issues, with the distress added to the pregnancy. Women with gestational diabetes experienced increased anxiety, depression, and stress. Not only does it affect mental health during pregnancy, but it also leads to an increased risk of postpartum depression. The risk is over 4 times greater than a normal pregnancy. It was found that physical activity could decrease the risk of postpartum depression. It is a form of therapy that can help reduce the stress of these mental health issues. The two main risks GDM imposes on the baby are growth abnormalities and chemical imbalances after birth, which may require admission to a neonatal intensive care unit. Infants born to mothers with GDM are at risk of being both large for gestational age (macrosomic) in unmanaged GDM, and small for gestational age and Intrauterine growth retardation in managed GDM. Macrosomia in turn increases the risk of instrumental deliveries (e.g., forceps, ventouse, and caesarean section) or problems during vaginal delivery (such as shoulder dystocia). Macrosomia may affect 12% of normal women compared to 20% of women with GDM.
It has been noted that the sequences translated from this locus and from that of β-actin are very similar to the predicted ones, suggesting a common ancestral sequence that suffered duplication and genetic conversion. In terms of pathology, it has been associated with processes such as amyloidosis, retinitis pigmentosa, infection mechanisms, kidney diseases, and various types of congenital hearing loss. Six autosomal-dominant point mutations in the sequence have been found to cause various types of hearing loss, particularly sensorineural hearing loss linked to the DFNA 20/26 locus. It seems that they affect the stereocilia of the ciliated cells present in the inner ear's Organ of Corti. β-actin is the most abundant protein found in human tissue, but it is not very abundant in ciliated cells, which explains the location of the pathology. On the other hand, it appears that the majority of these mutations affect the areas involved in linking with other proteins, particularly actomyosin. Some experiments have suggested that the pathological mechanism for this type of hearing loss relates to the F-actin in the mutations being more sensitive to cofilin than normal. However, although there is no record of any case, it is known that γ-actin is also expressed in skeletal muscles, and although it is present in small quantities, model organisms have shown that its absence can give rise to myopathies.

Unlike in multicellular organisms, increases in cell size (cell growth) and reproduction by cell division are tightly linked in unicellular organisms. Bacteria grow to a fixed size and then reproduce through binary fission, a form of asexual reproduction. Under optimal conditions, bacteria can grow and divide extremely rapidly, and some bacterial populations can double as quickly as every 17 minutes. In cell division, two identical clone daughter cells are produced. Some bacteria, while still reproducing asexually, form more complex reproductive structures that help disperse the newly formed daughter cells. Examples include fruiting body formation by myxobacteria and aerial hyphae formation by Streptomyces species, or budding. Budding involves a cell forming a protrusion that breaks away and produces a daughter cell. In the laboratory, bacteria are usually grown using solid or liquid media. Solid growth media, such as agar plates, are used to isolate pure cultures of a bacterial strain. However, liquid growth media are used when the measurement of growth or large volumes of cells are required. Growth in stirred liquid media occurs as an even cell suspension, making the cultures easy to divide and transfer, although isolating single bacteria from liquid media is difficult. The use of selective media (media with specific nutrients added or deficient, or with antibiotics added) can help identify specific organisms. Most laboratory techniques for growing bacteria use high levels of nutrients to produce large amounts of cells cheaply and quickly.
For example, SEC-MALS analysis of a monodisperse protein will show that the entire peak consists of molecules with the same molar mass, something that is not possible with standard SEC analysis. Determination of molar mass with SLS requires combining the light scattering measurements with concentration measurements. Therefore SEC-MALS typically includes the light scattering detector and either a differential refractometer or UV/Vis absorbance detector. In addition, MALS determines the rms radius Rg of molecules above a certain size limit, typically 10 nm. SEC-MALS can therefore analyze the conformation of polymers via the relationship of molar mass to Rg. For smaller molecules, either DLS or, more commonly, a differential viscometer is added to determine hydrodynamic radius and evaluate molecular conformation in the same manner. In SEC-DLS, the sizes of the macromolecules are measured as they elute into the flow cell of the DLS instrument from the size exclusion column set. The hydrodynamic size of the molecules or particles are measured and not their molecular weights. For proteins a Mark-Houwink type of calculation can be used to estimate the molecular weight from the hydrodynamic size. A major advantage of DLS coupled with SEC is the ability to obtain enhanced DLS resolution. Batch DLS is quick and simple and provides a direct measure of the average size, but the baseline resolution of DLS is a ratio of 3:1 in diameter. Using SEC, the proteins and protein oligomers are separated, allowing oligomeric resolution. Aggregation studies can also be done using ASEC.
=== Homeostasis === Copper is absorbed, transported, distributed, stored, and excreted in the body according to complex homeostatic processes which ensure a constant and sufficient supply of the micronutrient while simultaneously avoiding excess levels. If an insufficient amount of copper is ingested for a short period of time, copper stores in the liver will be depleted. Should this depletion continue, a copper health deficiency condition may develop. If too much copper is ingested, an excess condition can result. Both of these conditions, deficiency and excess, can lead to tissue injury and disease. However, due to homeostatic regulation, the human body is capable of balancing a wide range of copper intakes for the needs of healthy individuals. Many aspects of copper homeostasis are known at the molecular level. Copper's essentiality is due to its ability to act as an electron donor or acceptor as its oxidation state fluxes between Cu1+(cuprous) and Cu2+ (cupric). As a component of about a dozen cuproenzymes, copper is involved in key redox (i.e., oxidation-reduction) reactions in essential metabolic processes such as mitochondrial respiration, synthesis of melanin, and cross-linking of collagen. Copper is an integral part of the antioxidant enzyme copper-zinc superoxide dismutase, and has a role in iron homeostasis as a cofactor in ceruloplasmin. A list of some key copper-containing enzymes and their functions is summarized below:
Hadi's election on 24 February 2012 peacefully introduced a new government in Yemen, with only a small percentage of voters spoiling their ballots in the single-candidate contest. Hadi, a southerner, especially enjoyed support in former South Yemen, quieting the murmurs of separatism, although the Southern Movement boycotted the presidential election, as did the Houthis. Hadi did not give the restive Houthis any seats in his cabinet.

One of the most prominent campaigners was a young man from East Germany called Rudi Dutschke who also criticised the forms of capitalism that were to be seen in West Berlin. Just before Easter 1968, a young man tried to kill Dutschke as he bicycled to the student union, seriously injuring him. All over West Germany, thousands demonstrated against the Springer newspapers which were seen as the prime cause of the violence against students. Trucks carrying newspapers were set on fire and windows in office buildings broken. In the wake of these demonstrations, in which the question of America's role in Vietnam began to play a bigger role, came a desire among the students to find out more about the role of the parent-generation in the Nazi era. The proceedings of the War Crimes Tribunal at Nuremberg had been widely publicised in Germany; the attorney Fritz Bauer gathered evidence on the guards of the Auschwitz concentration camp, and about twenty were put on trial in Frankfurt in 1963. Daily newspaper reports and visits by school classes to the proceedings revealed to the German public the nature of the concentration camp system, and it became evident that the Shoah was of vastly greater dimensions than the German population had believed. The term "Holocaust" for the systematic mass-murder of Jews first came into use in 1979, when a 1978 American mini-series with that name was shown on West German television. The processes set in motion by the Auschwitz trial reverberated decades later.
β-carotene can be extracted from fungus Blakeslea trispora, marine algae Dunaliella salina or genetically modified yeast Saccharomyces cerevisiae, starting with xylose as a substrate. Chemical synthesis uses either a method developed by BASF or a Grignard reaction utilized by Hoffman-La Roche. The world market for synthetic retinol is primarily for animal feed, leaving approximately 13% for a combination of food, prescription medication and dietary supplement use. Industrial methods for the production of retinol rely on chemical synthesis. The first industrialized synthesis of retinol was achieved by the company Hoffmann-La Roche in 1947. In the following decades, eight other companies developed their own processes. β-ionone, synthesized from acetone, is the essential starting point for all industrial syntheses. Each process involves elongating the unsaturated carbon chain. Pure retinol is extremely sensitive to oxidization and is prepared and transported at low temperatures and oxygen-free atmospheres. When prepared as a dietary supplement or food additive, retinol is stabilized as the ester derivatives retinyl acetate or retinyl palmitate. Prior to 1999, three companies, Roche, BASF and Rhone-Poulenc controlled 96% of global vitamin A sales. In 2001, the European Commission imposed total fines of 855.22 million euros on these and five other companies for their participation in eight distinct market-sharing and price-fixing cartels that dated back to 1989. Roche sold its vitamin division to DSM in 2003. DSM and BASF have the major share of industrial production.
The War of 1812 coincided with the War of the Sixth Coalition. Historians in the United States and Canada see it as a war in its own right, while Europeans often see it as a minor theatre of the Napoleonic Wars. The United States declared war on Britain due to a variety of reasons, including British support for Native Americans resisting US settler expansion, interference with American merchant shipping, the impressment of Royal Navy deserters from US merchantmen, and expansionist American desires to occupy Canada. France had interfered with American shipping as well, and the United States had considered declaring war on France. The war ended in a military stalemate under the Treaty of Ghent, which took effect in early 1815 when Napoleon was still in Elba and returned the pre-war boundaries.
2 GSH + ROOH → GSSG + ROH + H2O Other enzymes, such as glutaredoxins, generate glutathione disulfide through thiol-disulfide exchange with protein disulfide bonds or other low molecular mass compounds, such as coenzyme A disulfide or dehydroascorbic acid.
=== General features === Although the tetrahedral intermediates are usually transient intermediates, many compounds of this general structures are known. The reactions of aldehydes, ketones, and their derivatives frequently have a detectable tetrahedral intermediate, while for the reactions of derivatives of carboxylic acids this is not the case. At the oxidation level of carboxylic acid derivatives, the groups such as OR, OAr, NR2, or Cl are conjugated with the carbonyl group, which means that addition to the carbonyl group is thermodynamically less favored than addition to corresponding aldehyde or ketone. Stable tetrahedral intermediates of carboxylic acid derivatives do exist and they usually possess at least one of the following four structural features:
