This is a working overview of triple agonist, written for readers who want more than a one-paragraph summary but less than a textbook.
Reviewed 2026-01-08. Anything still debated is marked as such rather than presented as settled.
Pharmacologically, retatrutide acts as a triple agonist at the glucagon-like peptide-1 receptor, the glucose-dependent insulinotropic polypeptide receptor, and the glucagon receptor. Activation of the first two receptors is associated with improved insulin secretion and reduced appetite. The glucagon receptor component is thought to increase energy expenditure, a mechanism that distinguishes this molecule from dual-agonist compounds. Researchers continue to investigate how the three activities interact and whether the combined profile offers advantages that justify additional clinical testing.
Several questions about the compound remain unresolved. The durability of weight reduction after treatment stops, the frequency of gastrointestinal side effects, and the long-term cardiovascular profile are topics of ongoing study. Regulatory submissions and phase 3 trial outcomes have not been fully reported in the public literature. Because most available data come from controlled trials rather than general-population use, conclusions about effectiveness outside study settings are provisional. The distinction between established findings and open questions matters when interpreting early coverage of the drug.

== Criticisms == In 2013, studies of autologous bone marrow stem cells on ventricular function were found to contain "hundreds" of discrepancies. Critics report that of 48 reports, just five underlying trials seemed to be used, and that in many cases whether they were randomized or merely observational accepter-versus-rejecter, was contradictory between reports of the same trial. One pair of reports of identical baseline characteristics and final results, was presented in two publications as, respectively, a 578-patient randomized trial and as a 391-subject observational study. Other reports required (impossible) negative standard deviations in subsets of people or contained fractional subjects, negative NYHA classes. Overall, many more people were reported as having receiving stem cells in trials, than the number of stem cells processed in the hospital's laboratory during that time. A university investigation, closed in 2012 without reporting, was reopened in July 2013. In 2014, a meta-analysis on stem cell therapy using bone-marrow stem cells for heart disease revealed discrepancies in published clinical trial reports, whereby studies with a higher number of discrepancies showed an increase in effect sizes. Another meta-analysis based on the intra-subject data of 12 randomized trials was unable to find any significant benefits of stem cell therapy on primary endpoints, such as major adverse events or increase in heart function measures, concluding there was no benefit.

The two discover that Tender has been running a round-tripping scheme, falsifying profits by overstating the costs of its acquisitions of third-party payment processors in Ghana and bribing officials to help stage publicity photos. Her findings enable Harper to make a public case against Tender at an investor conference, causing its stock to fall 28%. The Tender short is ultimately successful, netting the fund £110 million, with Sweetpea, Harper and Kwabena distributing £2 million amongst themselves. The three later explore an office for their fund.
Of these volunteers, 53 received daily active supplements containing meso-zeaxanthin, lutein, and zeaxanthin, while 52 subjects received a placebo (the control group). The outcome demonstrated that those receiving all three macular carotenoids had improved contrast sensitivity. The second trial, CREST AMD, was a two-year trial involving 96 subjects diagnosed with the early stages of AMD. All subjects received the AREDS2-recommended formula, with or without added meso-zeaxanthin, and all showed a significant improvement in the primary outcome measure of contrast sensitivity when reading an eye-chart. There was no difference between the results for subjects whose supplements included meso-zeaxanthin versus those who did not; thus, meso-zeaxanthin did not improve the eye health of the subjects who took it. There were no significant differences in how the subjects' AMD progressed, between the meso-zeaxanthin group and the AREDS2 group.
== Experimental setup == CV experiments are conducted on a solution in a cell fitted with electrodes. The solution consists of the solvent, in which is dissolved electrolyte and the species to be studied.

==== Reactions of tyrosines ==== Tyrosine residues are relatively unreactive; therefore they have not been a popular targets for bioconjugation. Recent development has shown that the tyrosine can be modified through electrophilic aromatic substitutions (EAS) reactions, and it is selective for the aromatic carbon adjacent to the phenolic hydroxyl group. This becomes particularly useful in the case that cysteine residues cannot be targeted. Specifically, diazonium effectively couples with tyrosine residues (diazonium salt shown as reagent in the first reaction in Figure 3 below), and an electron withdrawing substituent in the 4-position of diazonium salt can effectively increase the efficiency of the reaction. Cyclic diazodicarboxyamide derivative like 4-Phenyl-1,2,4-triazole-3,5-dione (PTAD) were reported for selective bioconjugation on tyrosine residues (the second reaction in Figure 3 below). A three-component Mannich-type reaction with aldehydes and anilines (the last reaction in Figure 3) was also described to be relatively tyrosine-selective under mild optimised reaction conditions.
In fluid dynamics, the continuity equation states that the rate at which mass enters a system is equal to the rate at which mass leaves the system plus the accumulation of mass within the system. The differential form of the continuity equation is:
== Function == In myocytes, sarcomeres adhere to the sarcolemma via costameres, which align at Z-discs and M-lines. The two primary cytoskeletal components of costameres are desmin intermediate filaments and gamma-actin microfilaments. It has been shown that gamma-actin interacting with another costameric protein dystrophin is critical for costameres forming mechanically strong links between the cytoskeleton and the sarcolemmal membrane. Additional studies have shown that gamma-actin colocalizes with alpha-actinin and GFP-labeled gamma actin localized to Z-discs, whereas GFP-alpha-actin localized to pointed ends of thin filaments, indicating that gamma actin specifically localizes to Z-discs in striated muscle cells. During development of myocytes, gamma actin is thought to play a role in the organization and assembly of developing sarcomeres, evidenced in part by its early colocalization with alpha-actinin. Gamma-actin is eventually replaced by sarcomeric alpha-actin isoforms, with low levels of gamma-actin persisting in adult myocytes which associate with Z-disc and costamere domains. Insights into the function of gamma-actin in muscle have come from studies employing transgenesis. In a skeletal muscle-specific knockout of gamma-actin in mice, these animals showed no detectable abnormalities in development; however, knockout mice showed muscle weakness and fiber necrosis, along with decreased isometric twitch force, disrupted intrafibrillar and interfibrillar connections among myocytes, and myopathy.
Lipid-based nanoparticles are very small spherical particles composed of lipids. They are a novel pharmaceutical drug delivery system (part of nanoparticle drug delivery), and a novel pharmaceutical formulation. There are many subclasses of lipid-based nanoparticles such as: lipid nanoparticles (LNPs), solid lipid nanoparticles (SLNs), and nanostructured lipid carriers (NLCs). Sometimes the term "LNP" describes all lipid-based nanoparticles. In specific applications, LNPs describe a specific type of lipid-based nanoparticle, such as the LNPs used for the mRNA vaccine. Using LNPs for drug delivery was first approved in 2018 for the siRNA drug Onpattro. LNPs became more widely known late in 2020, as some COVID-19 vaccines that use RNA vaccine technology coat the fragile mRNA strands with PEGylated lipid nanoparticles as their delivery vehicle (including both the Moderna and the Pfizer–BioNTech COVID-19 vaccines).

Initial letters are used where there is no ambiguity: C cysteine, H histidine, I isoleucine, M methionine, S serine, V valine. No other amino acids in this set begin with each of those letters. Where arbitrary assignment is needed, the structurally simpler amino acids are given precedence: A alanine, G glycine, L leucine, P proline, T threonine. For example, alanine is simpler than arginine or asparagine, the other amino acids starting with "a". F PHenylalanine and R aRginine are assigned by being phonetically suggestive, W tryptophan is assigned based on the double ring being visually suggestive to the bulky letter W, K lysine and Y tyrosine are assigned as alphabetically nearest to their initials L and T (note that U was avoided for its similarity with V, while X was reserved for undetermined or atypical amino acids); for tyrosine the mnemonic tYrosine was also proposed, D aspartate was assigned arbitrarily, with the proposed mnemonic asparDic acid; E glutamate was assigned in alphabetical sequence being larger by merely one methylene –CH2– group, N asparagine was assigned arbitrarily, with the proposed mnemonic asparagiNe; Q glutamine was assigned in alphabetical sequence of those still available (note again that O was avoided due to similarity with D), with the proposed mnemonic Qlutamine.
== Function == This gene encodes one of the two alpha chains of type XI collagen, a minor fibrillar collagen. It is located on chromosome 6 very close to but separate from the gene for retinoid X receptor beta. Type XI collagen is a heterotrimer but the third alpha chain is a post-translationally modified alpha 1 type II chain. Proteolytic processing of this type XI chain produces PARP, a proline/arginine-rich protein that is an amino terminal domain. Mutations in this gene are associated with type III Stickler syndrome, otospondylomegaepiphyseal dysplasia (OSMED syndrome), Weissenbacher-Zweymuller syndrome, and autosomal dominant nonsyndromic sensorineural 13 deafness. Three transcript variants encoding different isoforms have been identified for this gene.
== Applications == The products based on collagen proteins are used for food, health and nutrition, pharmaceutical industries and technical applications for example Photo, Ballistic, Restoration, Lubricants, Technical detergents.

It is designed as a triple agonist acting on the GLP-1, GIP, and glucagon receptors. This combination is intended to influence appetite, insulin secretion, and energy expenditure. Single-receptor and dual-receptor compounds act on a narrower set of targets.
No. It remains investigational, and phase 3 results have not been fully published or reviewed by regulators. Official approval status should be confirmed through regulatory agencies rather than secondary sources.
The added glucagon receptor activity is the main difference in its mechanism. Whether that addition produces meaningful benefits in clinical outcomes is still being studied. Comparisons between compounds rest largely on indirect rather than head-to-head trial data.
