Acting as a triple agonist, the molecule binds the GLP-1, GIP, and glucagon receptors. GLP-1 activity slows gastric emptying and dampens appetite, while GIP signaling contributes to insulin sensitivity and fat metabolism. Glucagon receptor engagement raises energy expenditure and encourages fat breakdown, although it can also elevate blood glucose. Combining three pathways is intended to yield larger weight reduction than single or dual agonists, and researchers continue to examine how the balance among them shapes tolerability.
Clinical studies have reported notable reductions in body weight among participants. Early trials measured safety and explored several dose levels, and later studies tracked body-weight change over months of treatment. Investigators also monitor effects on glycemic markers, liver fat, and blood lipids. Because the compound is still in development, questions about long-term safety, cardiovascular outcomes, and durability after treatment ends remain open.
Quantification of the peptide in biological samples generally relies on liquid chromatography coupled with tandem mass spectrometry. This approach separates the analyte from matrix components and detects it by mass-to-charge transitions specific to the molecule. Immunoassays offer higher throughput but can cross-react with related peptides and metabolites, so mass spectrometric methods are preferred when structural confirmation is required. Method validation typically addresses accuracy, precision, selectivity, and stability under handling conditions.
Several questions remain unresolved. It is not yet known whether the compound reduces cardiovascular events or mortality, because outcome studies require long follow-up. The durability of weight reduction after treatment withdrawal is uncertain, and rebound has been observed with other incretin-based therapies. Long-term safety data covering several years are limited. Effects in adolescents, in pregnancy, and in people with significant kidney or liver impairment have not been characterized in published reports.

Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.
Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
Receptor activation produces downstream effects that differ by tissue. GLP-1 receptor signaling influences appetite regulation and insulin secretion in a glucose-dependent manner. GIP receptor activity contributes to metabolic handling of nutrients and may modulate adipose tissue. Glucagon receptor engagement raises energy expenditure and promotes hepatic lipid turnover, though the balance among these actions in humans is still being characterized. Preclinical models showed reductions in body weight and improved glycemic markers.
Identification and purity assessment typically rely on reversed-phase high-performance liquid chromatography, often paired with mass spectrometry. Mass measurement confirms the expected molecular mass and can reveal truncations or modifications. Peptide mapping and sequencing techniques provide sequence-level confirmation when needed. Because related peptide impurities can behave similarly in a single method, orthogonal techniques are usually combined. Reported purity values depend heavily on the method used and should be interpreted with that in mind.
Dissolution behavior depends on the amino acid sequence, the counterion content, and the buffer chosen. Many peptides disperse readily in water or mild aqueous buffers, while others require a small amount of organic co-solvent or a change in pH. Adsorption to plastic and glass surfaces can reduce the concentration of a solution over time, particularly at low concentrations. Filtration before analysis removes particulates, and aliquoting limits repeated freeze-thaw cycles that stress the material.

Thalassemias are a group of inherited blood disorders that manifest as the production of reduced hemoglobin. Symptoms depend on the type of thalassemia and can vary from none to severe, including death. Often there is mild-to-severe anemia (low red blood cells or hemoglobin), as thalassemia can affect the production of red blood cells and also affect how long the red blood cells live. Symptoms include tiredness, pallor, bone problems, an enlarged spleen, jaundice, pulmonary hypertension, and dark urine. A child's growth and development may be slower than normal. Thalassemias are genetic disorders. Alpha thalassemia is caused by deficient production of the alpha globin component of hemoglobin, while beta thalassemia is a deficiency in the beta globin component. The severity of alpha and beta thalassemia depends on how many of the four genes for alpha globin or two genes for beta globin are faulty. Diagnosis is typically by blood tests including a complete blood count, special hemoglobin tests, and genetic tests. Diagnosis may occur before birth through prenatal testing. Treatment depends on the type and severity. Clinically, thalassemia is classed as Transfusion-Dependent Thalassemia (TDT) or non-Transfusion-Dependent Thalassemia (NTDT), since this determines the principal treatment options. TDT requires regular blood transfusions, typically every two to five weeks. TDTs include beta-thalassemia major, hemoglobin H disease, and severe HbE/beta-thalassemia. NTDT does not need regular transfusions but may require transfusion in case of an anemia crisis.
== Contraindications == Bicalutamide is pregnancy category X, or "contraindicated in pregnancy", in the U.S., and pregnancy category D, the second most restricted rating, in Australia. As such, it is contraindicated in women during pregnancy, and women who are sexually active and who can or may become pregnant are strongly recommended to take bicalutamide only in combination with adequate contraception. It is unknown whether bicalutamide is excreted in breast milk, but many drugs are excreted in breast milk, and for this reason, bicalutamide treatment is similarly not recommended while breastfeeding. In individuals with severe, though not mild-to-moderate hepatic impairment, there is evidence that the elimination of bicalutamide is slowed, and hence, caution may be warranted in these patients as circulating levels of bicalutamide may be increased. In severe hepatic impairment, the elimination half-life of the active (R)-enantiomer of bicalutamide is increased by about 1.75-fold (76% increase; elimination half-life of 5.9 and 10.4 days for normal and impaired patients, respectively). The elimination half-life of bicalutamide is unchanged in renal impairment.

Intensity-fading MALDI is a term coined to rename an existing method originally reported in 1999 to indirectly study a Protein–protein interaction or other protein complex and was the same year applied to a biological mixture to study the antigenicity of the influenza virus. It involves treating a protein and a potential binding partner with a site-specific endoproteinase with the binding sites identified by their reduced area (or intensity) in a MALDI mass spectrum compared to that of non-bound protein control. It was falsely reported as new and novel in a later application by a Spanish group. The true origins of the approach and a range of applications including those employing gel based separations, drug-protein interactions and the relative affinity of such interactions, are described in a review article.
=== Birth of the supermarket === Historically, there has been much debate about the origin of the supermarket. For example, Southern California grocery store chains Alpha Beta and Ralphs both have strong claims to being the first supermarket. By 1930, both chains were already operating multiple 12,000-square-foot (1,100 m2) self-service grocery stores. However, as of 1930, both chains were not yet true supermarkets in the modern sense because their prices remained quite high; as noted above, one of the most important defining features of the supermarket is cheap food. Their main selling point was free parking. Other strong contenders in Texas included Weingarten's and Henke & Pillot. To end the debate, the Food Marketing Institute in conjunction with the Smithsonian Institution and with funding from H.J. Heinz, researched the issue. They defined the attributes of a supermarket as "self-service, separate product departments, discount pricing, marketing and volume selling". They determined that the first true supermarket in the United States was opened by a former Kroger employee, Michael J. Cullen, on 4 August 1930, inside a 6,000-square-foot (560 m2) former garage in Jamaica, Queens in New York City. The store King Kullen, operated under the logic of "pile it high and sell it cheap". The store layout was designed by Joseph Unger, who originated the concept of customers using baskets to collect groceries before checking out at a counter. Everything displayed for sale in the store "had prices clearly marked", meaning that consumers would no longer need to haggle over prices.
February 23, 2011 Venezuela Economists expressed concern with regards to Venezuela's economy improving due to instability in the private sector pressured by the socialist government, as José Guerra, former manager at the Central Bank of Venezuela, stated that private investment, accounting for half of Venezuela's GDP, dropped 2.2% in 2010.

== Advantages and disadvantages == Advantages include minimal overlap in metal signals meaning the instrument is theoretically capable of detecting 100 parameters per cell, entire cell signaling networks can be inferred organically without reliance on prior knowledge, and one well-constructed experiment produces large amounts of data. Disadvantages, in the case of CyTOF, include the practical flow rate is around 500 cells per second versus several thousand in flow cytometry and current reagents available limit cytometer use to around 50 parameters per cell. Additionally, mass cytometry is a destructive method and cells cannot be sorted for further analysis. In the case of IMC, the resolution of the data is relatively low (1μm2/pixel), the technique is as well destructive, acquiring of the data is also very slow, and it requires specialized expensive equipment and expertise.

Through this catabolic process, GSNOR regulates the cellular concentrations of GSNO and plays a central role in regulating the levels of endogenous S-nitrosothiols and controlling protein S-nitrosylation-based signaling. As an example of S-nitrosylation-based signaling, Barglow et al. showed that GSNO selectively S-nitrosylates reduced thioredoxin at cysteine 62. Nitrosylated thioredoxin, via directed protein-protein interaction, trans-nitrosylates the active site cysteine of caspase-3 thus inactivating caspase-3 and preventing induction of apoptosis. As might be expected of an enzyme involved in regulating NO levels and signaling, pleiotropic effects are observed in GSNOR knockout models. Deleting the GSNOR gene from both yeast and mice increased the cellular levels of GSNO and nitrosylated proteins, and the yeast cells showed increased susceptibility to nitrosative stress. Null mice show increased levels of S-nitrosated proteins, increased beta adrenergic receptor numbers in lung and heart, diminished tachyphylaxis to β2-adrenergic receptor agonists, hyporesponsiveness to methacholine and allergen challenge and reduced infarct size after occlusion of the coronary artery. In addition, null mice show increased tissue damage and mortality following challenge with bacteria or endotoxin and are hypotensive under anesthesia yet normotensive in the conscious state.
=== Artificial DNA nanostructures === The success of DNA nanotechnology in constructing artificially designed nanostructures out of nucleic acids such as DNA, combined with the demonstration of systems for DNA computing, has led to speculation that artificial nucleic acid nanodevices can be used to target drug delivery based upon directly sensing its environment. These methods make use of DNA solely as a structural material and a chemical, and do not make use of its biological role as the carrier of genetic information. Nucleic acid logic circuits that could potentially be used as the core of a system that releases a drug only in response to a stimulus such as a specific mRNA have been demonstrated. In addition, a DNA "box" with a controllable lid has been synthesized using the DNA origami method. This structure could encapsulate a drug in its closed state, and open to release it only in response to a desired stimulus.
==== Surfactants ==== The purpose of surfactants is to mobilize various components of NAPLs by lowering their viscosity and interfacial tension. Solubilizing agents increase the solubility of NAPLs and transfer it to the aqueous phase, allowing it to then be extracted and treated. Mobilizing agents target the residually saturated component of NAPL, allowing it to be displaced by continuous flooding. While surfactants are highly effective, resulting in recovery of 94% of the original DNAPL in case studies, they are also expensive and cost-prohibitive, also potentially adversely affecting the pH of the subsurface environment.
=== Tunnelling currents DNA sequencing === Another approach uses measurements of the electrical tunnelling currents across single-strand DNA as it moves through a channel. Depending on its electronic structure, each base affects the tunnelling current differently, allowing differentiation between different bases. The use of tunnelling currents has the potential to sequence orders of magnitude faster than ionic current methods and the sequencing of several DNA oligomers and micro-RNA has already been achieved.

It is a synthetic peptide classified as a triple receptor agonist. It engages the GLP-1, GIP, and glucagon receptors at once. It is investigated for metabolic and weight-related conditions rather than approved for general use.
No regulatory agency has approved it for routine medical use. It remains an investigational compound studied in controlled trials. Access outside research settings is not established.
Each receptor influences energy balance through different pathways. Combining them is intended to add fat loss from lowered intake and higher expenditure. The optimal balance among the three effects is still under study.
Trials measure percentage change in body weight, absolute weight loss, and glycemic markers such as hemoglobin A1c. They also record blood pressure, lipids, and liver fat. Adverse events are tracked throughout.
